Invitrogen™ S.N.A.P.™ UV-Free Gel Purification Kit
Description
Includes
Each S.N.A.P. UV-Free Gel Purification Kit includes enough crystal violet reagents, S.N.A.P. columns, collection vials, sodium iodide solution, purification buffers, and sterile water for 25 gel purifications.
The S.N.A.P.™ UV-Free Gel Purification Kit uses non-toxic reagents to rapidly purify intact DNA fragments from agarose gels. The key to isolating undamaged DNA is the unique crystal violet reagent, which allows visualization of DNA (≥200 ng) in the gel under ambient light. This eliminates the need for ethidium bromide and exposure to UV light, which can nick and damage DNA. Nucleic acids up to 7 kb are efficiently purified from the gel using the convenient S.N.A.P.™ resin, provided ready to use in a spin column format. The resin eliminates organic extractions, which generate hazardous waste. In addition, there are no time-consuming ethanol precipitations.
Easy to Use
The S.N.A.P.™ UV-Free Gel Purification Kit yields pure, intact DNA in an easy-to-perform, rapid procedure. Simply:
1. Prepare gel with crystal violet reagent and load samples in crystal violet loading buffer.
2. Visualize DNA band under ambient light and excise from agarose gel.
3. Add NaI solution (provided) and melt the agarose at 42-50°C.
4. Load sample onto the S.N.A.P.™ spin column.
5. Wash away contaminants.
6. Elute DNA in water.
The eluted DNA is highly pure, intact, and ready to use for cloning, sequencing, transformation, or transfection.

Frequently Asked Questions (FAQs)
Yes, both TAE and TBE agarose gels are compatible with our gel extraction kits.
No, TBE gels contain borate, which interferes with the sodium iodide solution. Therefore, only TAE gels can be used. We recommend the PureLink Quick Gel Extraction kit for both TAE and TBE agarose gels.
S.N.A.P. mini prep columns have been used to purify fragments of 100 bp in gel purification procedures.
The column contains a silica/glass fiber based resin. However, the exact composition is proprietary.
The S.N.A.P. UV-free Gel Purification Kit utilizes crystal violet as a staining reagent, which can be visualized in non-UV light conditions. This can prevent DNA damage that can be caused by the UV light required by normal ethidium bromide staining. DNA fragments recovered using this kit have higher cloning efficiency. In addition, DNA can be visualized under normal light as a thin violet band while the gel is running and excised as soon as bands are sufficiently resolved.
