Kasugamycin (Ksg) hydrochloride hydrate is an antibiotic that binds to 30s and 70s ribosomes, but not to the 50s subunit, and exhibits anti-infective activity. It mimics mRNA nucleotides, disrupting tRNA binding and inhibiting canonical translation initiation. It increases the sensitivity of mycobacteria to Rifampicin in vitro and in mouse infection models.
- Inhibits P-site tRNA binding by destabilizing mRNA.
- Decreases the mistranslation rates of asparagine-to-aspartate and glutamine-to-glutamate.
- Reduces mycobacterial rifampicin phenotypic resistance in wild-type and high mistranslating gatA mutant mycobacterial strains.
- Results in dose-dependent concentration-time profiles in plasma with rapid clearance of M. tuberculosis in infected mice.
- Increases Rifampicin susceptibility by 30-fold in mouse models.
