Kavain is a class of kavalactone isolated from *Piper methysticum*, which has anxiolytic properties in animals and humans. It positively modulates γ-Aminobutyric acid type A (GABAA) receptor. In vitro, Kavain positively modulates all GABAA receptors regardless of subunit composition, with a greater enhancement at α4β2δ than at α1β2γ2L GABAARs. Its modulatory effect is not affected by flumazenil, indicating it does not enhance GABAARs via the classical benzodiazepine binding site.
