Kligler Iron Agar (KIA) is a differential culture medium used for the identification of Gram negative enteric bacteria based on carbohydrate fermentation and hydrogen sulfide production. Each tube contains approximately 9mL of agar prepared as a slant with a deep butt, creating aerobic and anaerobic reaction zones. The formulation contains two carbohydrates, lactose and glucose, along with the pH indicator phenol red. Organisms that ferment only glucose produce limited acid that turns the butt yellow while the slant reverts to red as proteins are metabolized. Organisms that ferment both glucose and lactose produce larger amounts of acid, turning both the slant and butt yellow. The medium also contains iron salts that react with hydrogen sulfide to form a black precipitate, indicating sulfur reduction.
