LB Agar (Lennox) with Ampicillin is a convenient pre mixed selective medium widely used in molecular biology for isolating plasmid transformed Escherichia coli. Each plate contains 20 mL of Lennox formulation Lysogeny Broth agar (5 g/L NaCl) supplemented with 100 µg/mL Ampicillin. This ready to use format eliminates the time consuming and temperature sensitive process of adding antibiotics to molten agar. The 100 µg/mL concentration is the standard for maintaining high copy number plasmids carrying the bla gene (beta lactamase). Ampicillin inhibits cell wall synthesis, preventing the growth of non resistant cells, while transformed bacteria degrade the antibiotic and form colonies. The Lennox low salt formulation is beneficial for salt sensitive downstream applications and routine cloning workflows.
