LB Agar (Miller) with Ampicillin and Chloramphenicol is a dual selection medium designed for molecular biology workflows requiring maintenance of two resistance markers. Each plate contains 20 mL of high salt Miller formulation Lysogeny Broth agar supplemented with both antibiotics. This medium is commonly used in co transformation experiments where a single E. coli cell must maintain two compatible plasmids, such as an expression vector and a reporter plasmid. It is also suitable for specialized strains like BL21(DE3)pLysS or Rosetta that carry helper plasmids conferring chloramphenicol resistance. Ampicillin maintains plasmid selection while chloramphenicol ensures retention of the helper plasmid, allowing stable propagation of both genetic elements during cloning and protein expression experiments.
