The UbiTest kit determines whether a target protein is polyubiquitinated and improves on traditional immunoprecipitation–Western blot methods, which often fail due to epitope masking, reduced antibody affinity, or altered selectivity. By combining immunoprecipitation with broad‑spectrum DUB digestion, the assay reveals increased signal for the protein of interest after DUB treatment, confirming ubiquitination even when untreated samples show weak or no detection. To avoid antibody‑dependent variability, UbiTest uses TUBEs to pull down polyubiquitinated proteins, leveraging their nanomolar affinity and ability to bind all eight linkage types. For linkage determination, LifeSensors employs linkage‑specific DUBs; an increased unmodified band after K48‑ or K63‑specific DUB treatment indicates the protein carries those linkage types.
