This assay kit employs a liposome-encapsulated dye to monitor dye release due to lipid bilayer damage. It screens compounds that disrupt permeability and damage cell membranes, such as pore-forming cytotoxins. Fluorescence is monitored at 580 nm (excitation at 540 nm) in a high-throughput 96-well plate format.
- Assay for screening compound collections.
- Liposomes composed of HSPC: cholesterol: DSPE-PEG2000 (3:1:1 mass ratio).
- Encapsulated dye is rhodamine conjugated with diethylenetriamine, quenched inside the liposome.
- Released dye generates fluorescence in the presence of a color development agent.
- Includes 1.25 mL of liposomal dye, 1.25 mL of color development solution, 7.5 mL of buffer, and 1 mL of DMSO.
- Designed for 100 samples using a 96-well plate.
- Protocol provided for 96-well plate fluorescence plate reader with a total sample volume of 100 μL.
