Litmus Milk Medium is a classic differential medium used to characterize bacteria based on their metabolic activity toward milk components such as lactose and casein. Each tube contains 10 mL of skim milk supplemented with the litmus indicator, which functions as both a pH and redox indicator. The medium allows multiple biochemical reactions to be observed in a single test, supporting identification of organisms such as Clostridium, Enterococcus, and Lactobacillus. Lactose fermentation produces acid that turns the medium pink, while proteolysis releases alkaline products that shift the color toward blue or purple. Additional reactions include curd formation from casein coagulation, peptonization producing a clear fluid layer, and reduction reactions where the indicator becomes colorless under anaerobic conditions.
