Lometrexol (DDATHF) is an antipurine antifolate that inhibits glycinamide ribonucleotide formyltransferase (GARFT) and de novo purine synthesis. It causes abnormal cell proliferation, apoptosis, and cell cycle arrest. Lometrexol exhibits anticancer activity and is a potent human Serine hydroxymethyltransferase1/2 (hSHMT1/2) inhibitor. It binds tightly to GART, depleting intracellular purine ribonucleotides.
