M Endo LES Agar is a selective and differential medium used for the enumeration of total coliform bacteria in water samples using the membrane filtration method. LES stands for Lawrence Experimental Station, indicating a formulation optimized for improved coliform recovery. Each plate contains approximately 20 mL of distinctive reddish pink agar designed to differentiate bacteria based on lactose fermentation. The medium includes sodium sulfite and basic fuchsin, which inhibit Gram positive organisms while acting as indicators. When membrane filters containing bacteria are placed on the agar surface, lactose fermenting coliforms such as E. coli produce red colonies often displaying a characteristic green metallic sheen. Non lactose fermenters appear colorless or pale pink, allowing reliable identification of coliform contamination in water samples.
