m-PEG5-azide is a PEG-based PROTAC linker used in the synthesis of PROTACs. It functions as a click chemistry reagent, containing an Azide group. It can participate in copper-catalyzed azide-alkyne cycloaddition (CuAAc) reactions with molecules containing Alkyne groups, and also strain-promoted alkyne-azide cycloaddition (SPAAC) reactions with molecules containing DBCO or BCN groups. PROTACs themselves contain two distinct ligands connected by a linker; one targets an E3 ubiquitin ligase and the other targets the desired protein, leveraging the intracellular ubiquitin-proteasome system for selective protein degradation.
