MacConkey Broth with a 90 mL fill volume is specifically designed for preparing accurate 1:10 sample dilutions in microbiological testing. Laboratories can directly add a 10 g or 10 mL sample to the bottle to achieve a final volume of 100 mL, simplifying dilution preparation. The medium functions as a selective enrichment broth for detection of lactose fermenting coliform bacteria. Bile salts inhibit Gram positive organisms, allowing Gram negative enteric bacteria to grow. Lactose serves as the fermentable substrate, and bromocresol purple is commonly used as the pH indicator. When coliforms ferment lactose they produce acid and gas, turning the medium from purple to yellow. If no fermentation occurs the broth remains purple, indicating absence of coliform contamination.
