MacConkey Inositol Carbenicillin Agar (MICA) is a specialized selective medium developed for the isolation and identification of Klebsiella species, particularly Klebsiella pneumoniae, from mixed microbial samples. This formulation modifies standard MacConkey agar in two key ways. Lactose is replaced with inositol as the fermentable carbohydrate, which Klebsiella species readily metabolize to produce acid. The medium is also supplemented with the antibiotic carbenicillin, which suppresses many competing Gram negative and Gram positive organisms. When Klebsiella ferments inositol, colonies appear pink to red due to acid production and often exhibit a characteristic mucoid appearance from capsule formation. These features allow reliable differentiation of Klebsiella from other organisms present in clinical or environmental samples.
