Mannitol Salt Agar (MSA), USP, is a selective and differential microbiological medium used for the isolation and differentiation of Staphylococcus species, particularly Staphylococcus aureus. The high sodium chloride concentration of approximately 7.5% inhibits the growth of many non halophilic organisms, making the medium highly selective for staphylococci. Mannitol serves as a fermentable carbohydrate, while phenol red acts as a pH indicator to differentiate fermenting and non fermenting organisms. Staphylococcus aureus ferments mannitol and produces acid that changes the medium from red to yellow, whereas non fermenting species maintain the original color. Each plate contains approximately 20mL of agar in mono packaging, suitable for routine microbiological testing. These plates are widely used in pharmaceutical quality control, food safety laboratories, and environmental monitoring programs.
