This gene encodes MEKK4 (also known as MTK1) a MAPK kinase kinase (MAPKKK) that activates stress-responsive MAPK pathways MAPK cascades consist of three kinases a MAPKKK activates a MAPKK which then activates a MAPK MEKK4 specifically activates the CSBP2 (p38) and JNK pathways but not the ERK pathway It contains a kinase domain at the C-terminus and a potential regulatory domain at the N-terminus MEKK4 is activated by environmental stresses such as UV radiation osmotic shock and inflammatory signals In vitro MEKK4 phosphorylates and activates PRKMK6 and SERK1 MAPKKs for CSBP2 and JNK respectively but not PRKMK1 which activates ERKs MEKK4 is a key mediator of CSBP2 activation and a minor contributor to JNK signaling Multiple alternatively spliced isoforms of MEKK4 have been identified
