Methyl Red–Voges Proskauer (MR VP) Broth is a dual purpose biochemical medium used for differentiating members of the Enterobacteriaceae family based on their glucose fermentation pathways. Each tube contains approximately 10 mL of buffered peptone glucose broth designed to support bacterial growth during incubation. After growth, the culture is divided to perform two tests. The Methyl Red test detects mixed acid fermentation, producing a stable acidic end product that turns the methyl red indicator bright red. The Voges Proskauer test detects acetoin, a neutral fermentation intermediate, through the addition of alpha naphthol and potassium hydroxide reagents. These complementary reactions help distinguish organisms such as Escherichia coli, Enterobacter, and Klebsiella during biochemical identification.
