Minimal Glucose Agar, commonly referred to as MTO or M9 Minimal Medium, is a chemically defined culture medium used for cultivation of prototrophic bacteria and for studying nutritional requirements of auxotrophic strains. Each plate contains approximately 30 mL of agar, providing extra depth that helps prevent dehydration during extended incubation periods. Unlike complex media such as LB or Nutrient Agar that contain undefined biological extracts, this medium includes only defined components. Glucose serves as the primary carbon and energy source, while inorganic salts supply nitrogen, magnesium, and buffering capacity. Because the exact chemical composition is known, Minimal Glucose Agar is widely used in bacterial genetics experiments, metabolic studies, and selection assays involving genetically engineered strains.
