MK2-IN-5 acetate (Hsp25 kinase inhibitor acetate) is a Mk2 pseudosubstrate with a Ki of 8 μM. It targets the protein interaction domain in the MAPK pathway and inhibits HSP25 and HSP27 phosphorylation. In vitro, MK2-IN-5 acetate (5-10 μM, 2 hours) inhibits TGF-β1 induced connective tissue growth factor (CTGF) and collagen type I in primary human keloid fibroblasts. In vivo, a single intraperitoneal injection of 2 mg/kg of MK2-IN-5 acetate decreased HSP25 phosphorylation in a ventilator-associated lung injury model.
