MLN120B dihydrochloride is a potent, orally active IKKβ inhibitor (IC50 60 nM) that inhibits multiple myeloma cell growth in vitro and in vivo. It is also used for rheumatoid arthritis research. In vitro, it blocks IκB phosphorylation/degradation and TNF-a-induced p65 NF-κB phosphorylation, reducing proliferation in various multiple myeloma cell lines. In vivo, oral administration reduces tumor growth markers and protects against bone and cartilage destruction in rheumatoid arthritis models.
