Modified Edwards Agar with Bovine Plasma is a selective and differential medium designed for the isolation and identification of Streptococcus species associated with bovine mastitis, particularly Streptococcus agalactiae. The formulation contains selective agents such as crystal violet and thallium sulfate that suppress contaminating organisms commonly present in raw milk, allowing streptococci to grow more clearly. Differentiation is primarily based on esculin hydrolysis. Organisms capable of hydrolyzing esculin react with ferric ions in the medium to form dark brown or black colonies, while Streptococcus agalactiae typically remains blue or colorless. The addition of bovine plasma enriches the medium and enhances characteristic colony appearance, supporting reliable screening of dairy samples in veterinary and food microbiology laboratories.
