Modified m TEC Agar is a chromogenic medium developed for the detection and enumeration of Escherichia coli in water samples using the membrane filtration technique. Each plate contains approximately 4.3 mL of agar and is designed to accommodate standard 47 mm membrane filters used in water quality testing. The formulation follows EPA Method 1603 and simplifies the traditional m TEC procedure by eliminating the need for transfer to a secondary substrate. The medium incorporates a chromogenic substrate that reacts with the enzyme β glucuronidase produced by most E. coli strains. When the substrate is cleaved, colonies develop a distinctive red to magenta coloration, allowing easy identification. Non target organisms generally appear colorless or pale, enabling reliable enumeration in environmental monitoring programs.
