Moeller Decarboxylase Broth is a differential biochemical medium used to determine whether bacteria produce decarboxylase enzymes that act on specific amino acids. This test is widely used for the identification and differentiation of Gram negative enteric bacilli, including members of the Enterobacteriaceae family. Each 5 mL tube contains glucose, peptones, and pH indicators such as bromocresol purple and cresol red that visually demonstrate metabolic activity. Following inoculation, organisms initially ferment glucose, producing acid and turning the medium yellow. If the organism produces a decarboxylase enzyme, alkaline amines are formed that raise the pH and restore the medium to purple. Organisms lacking the enzyme keep the medium acidic and yellow. This reaction pattern provides a reliable biochemical tool for organism differentiation.
