Moeller Decarboxylase Broth with Ornithine is a biochemical medium used to determine whether microorganisms produce the enzyme ornithine decarboxylase. The test is frequently used for differentiation of Gram negative enteric bacteria such as Enterobacter, Klebsiella, and Proteus species. Each 5 mL tube contains glucose, peptones, the amino acid ornithine, and pH indicators including bromocresol purple and cresol red. After inoculation, organisms ferment glucose and create an acidic environment that turns the medium yellow. If the organism possesses ornithine decarboxylase, the enzyme converts ornithine into the alkaline compound putrescine. This raises the pH and changes the medium back to purple. When the enzyme is absent, the medium remains yellow, indicating a negative reaction useful for biochemical identification.
