Motility Indole Ornithine (MIO) Medium is a semi solid, three in one diagnostic tube used to differentiate members of the Enterobacteriaceae family. The medium simultaneously evaluates motility, indole production, and ornithine decarboxylase activity, allowing efficient identification of organisms such as Klebsiella, Enterobacter, and Escherichia coli. The tube is inoculated with a straight stab through the center of the medium. Motility is indicated by turbidity diffusing outward from the stab line, while non motile organisms grow only along the stab path. Ornithine decarboxylation is detected by a shift of the bromocresol purple indicator from yellow back to purple as alkaline amines are produced. Indole production is determined after incubation by adding Kovac’s reagent, where a red layer indicates a positive reaction.
