Mueller Hinton Agar with 5% Sheep Blood is the internationally recognized standard medium for antimicrobial susceptibility testing of fastidious bacteria using the Kirby Bauer disk diffusion method. These plates are supplied in a large 150 mm format containing approximately 75 mL of agar, ensuring uniform depth and optimal diffusion of antimicrobial agents. While standard Mueller Hinton Agar supports many rapidly growing bacteria, fastidious organisms such as Streptococcus pneumoniae and Neisseria species require additional growth factors. The addition of 5% defibrinated sheep blood provides essential nutrients including hemin and NAD while also enabling observation of hemolytic reactions. The large plate diameter allows laboratories to test multiple antibiotic disks simultaneously with accurate and reproducible results.
