Nimustine hydrochloride (ACNU) is the hydrochloride salt form of Nimustine. This alkylating agent induces DNA double-strand breaks (DSBs) and inter-strand crosslinks (ICLs), activating the DNA damage response (DDR) signaling pathway. It also activates the p38 MAPK/JNK signaling pathway and demonstrates antitumor activity.
- Exhibits cytotoxicity against human U87MG cells with an IC50 of 0.72 μM.
- Activates caspase and AP-1, upregulates bim expression, and promotes c-Jun phosphorylation in LN-229 cells.
- Induces apoptosis and increases expressions of cleaved bim and caspase-3/8/9 in LN-229 cells.
- Inhibits tumor growth in C3H/HeN mouse models when combined with radiotherapy.
