Nitrate Broth is a differential medium used to determine whether microorganisms can reduce nitrate to nitrite or further nitrogenous compounds through nitrate reductase activity. This biochemical test is commonly used for identification of Gram negative bacteria including Enterobacteriaceae, Pseudomonas, and certain Mycobacterium species. Each 5 mL tube contains nutrient broth supplemented with potassium nitrate. After incubation, reagents containing sulfanilic acid and alpha naphthylamine are added to detect nitrite production. A red color indicates a positive nitrate reduction result. If no color appears, zinc dust may be added as a confirmation step to determine whether nitrate remains unreduced or has been reduced beyond nitrite. The test provides valuable metabolic information for bacterial identification.
