NucleoSpin RNA Blood provides a simple and convenient method for purifying RNA from up to 400 µl of fresh or frozen whole blood that has been stabilized using anticoagulants such as EDTA, citrate, or heparin. RNA is isolated via direct total blood lysis without the need for selective erythrocyte lysis at 4 °C, such that the entire procedure is performed at room temperature. Following binding of RNA to NucleoSpin RNA Blood mini spin columns, DNA is efficiently removed via an on-column rDNase digestion. Contaminants and inhibitors are then removed with two different wash buffers, and RNA is eluted with RNase-free water. The highly efficient kit protocol enables processing of 6 samples in less than an hour.
