NucView 488 Caspase-3 substrate is a novel, fluorogenic substrate for real-time or endpoint detection of apoptosis in live cells. The non-toxic, no-wash substrate is non-inhibitory to caspase-3 unlike other caspase substrates or FLICA assays permitting multiday analysis. Consisting of a high-affinity DNA dye linked to a DEVD caspase-3 substrate moiety, the initially non-fluorescent, cell permeable substrate is cleaved by caspase-3 in apoptotic cells and emits green fluorescence for the detection of both caspase-3 activity and apopototic nuclear morphology, in single-cells. As apoptotic rates and expression of asociated markers varies between cells, even within the same population, single-cell detection is important. Suitable for flow cytometry, fluorescence microscopy, live cell imaging systems, or fluorescence microplate reader, staining is compatible with fixation and permeabilization for immunostaining. NucView 488 is validated in over 100 cell types and 200 publications.
