This gene encodes a 186 kDa precursor protein that is part of the nucleoporin family and plays a key role in nuclear pore complexes (NPCs) which regulate macromolecule transport between the nucleus and cytoplasm The precursor undergoes autoproteolytic cleavage to generate two nucleoporins a 98 kDa nucleoporin containing a Gly-Leu-Phe-Gly (GLGF) repeat domain involved in nuclear import export mitotic progression and gene expression regulation and a 96 kDa nucleoporin which serves as a scaffold component of the NPC Proteolytic cleavage is crucial for targeting these proteins to the NPC Gene translocations have been linked to various leukemias creating chimeras with the GLGF domain fused to partner genes Alternative splicing results in multiple isoforms some lacking the 96 kDa nucleoporin region
