OF (Oxidation Fermentation) Medium Agar with Dextrose is a differential medium used to determine whether Gram negative bacteria metabolize carbohydrates through oxidation or fermentation. The medium contains glucose as the carbohydrate source and bromothymol blue as the pH indicator. In practice, the test is performed using two tubes for each organism. One tube remains open to air to detect oxidative metabolism, while the second tube is sealed with sterile mineral oil to create anaerobic conditions for fermentation. Acid production changes the indicator from green to yellow. Oxidative organisms produce acid only in the open tube, while fermentative organisms produce acid in both tubes. This test is widely used in microbiology laboratories for differentiating non enteric Gram negative rods and identifying organisms such as Pseudomonas and Enterobacteriaceae.
