Oligomycin Complex is a mixture composed primarily of Oligomycin A B and C It functions as a mitochondrial ATP synthase inhibitor targeting specifically the F0 subunit to disrupt oxidative phosphorylation via inhibition of ATP generation Oligomycin A exhibits selective inhibition of ATP synthase with reported GI50 at approximately 270 nM By blocking mitochondrial ATP synthesis oligomycin promotes cellular metabolic shift from oxidative phosphorylation to glycolysis In cell culture studies using various cancer cell lines oligomycin treatment has been demonstrated to suppress mitochondrial respiration and can induce apoptosis Additionally oligomycin has been employed in research to examine cellular bioenergetics adaptations mitochondrial-related apoptosis pathways and synergy of chemotherapeutic combinations in vitro
