Orange Serum Agar (OSA) is a specialized medium developed for the cultivation, isolation, and enumeration of aciduric spoilage microorganisms commonly associated with citrus and fruit juice products. The formulation includes components derived from orange serum that simulate the natural nutrient environment of fruit beverages. The medium is adjusted to an acidic pH of approximately 5.5, which inhibits many non spoilage bacteria while allowing acid tolerant microorganisms to grow. Nutrients such as yeast extract and tryptone support the development of organisms including lactic acid bacteria, yeasts, and molds that can impact product shelf life. OSA is widely used as a quality control medium in beverage processing laboratories for monitoring microbial contamination in juices and fruit based drinks.
