PD-1 also known as programmed cell death protein-1 is a cell surface receptor that belongs to the CD28 immunoglobulin super family and is expressed on activated T cells, B cells and NK cells. Studies have shown that PD-1/PD-L interaction function as an immune checkpoint for induction and maintenance of T-cells involved in peripheral tolerance and protects tissues from autoimmune attack. PD-1 down regulates the immune system which in turn reduces autoimmunity and promotes self-tolerance. In a tumor microenvironment, cancer cells utilize this PD-1/PD-L1 mechanism of immune suppression which confers survival advantage to the cancer cells.
