Phenol Red Broth with Maltose and Durham Tube is a differential test medium used to determine whether microorganisms can ferment the carbohydrate maltose. The broth contains maltose as the sole fermentable sugar and phenol red as the pH indicator. When an organism ferments maltose, acid is produced and the medium changes color from red to yellow. If fermentation also produces gas, the gas collects in the inverted Durham tube placed inside the broth. Lack of fermentation leaves the broth red, while alkaline reactions may produce a pink coloration. This test is widely used in microbiology laboratories for the identification and differentiation of Gram negative enteric bacteria and other microorganisms based on their carbohydrate fermentation patterns.
