Phenylalanine Agar Slant is a differential medium used to detect the presence of the enzyme phenylalanine deaminase in certain Gram negative bacteria. The medium contains the amino acid phenylalanine as the substrate. Organisms that produce phenylalanine deaminase convert phenylalanine into phenylpyruvic acid during incubation. After growth, a few drops of ferric chloride reagent are added to the surface of the slant. If phenylpyruvic acid is present, it reacts with the ferric chloride to produce a distinctive green coloration, indicating a positive test result. This reaction is commonly used in microbiology laboratories to differentiate members of the Enterobacteriaceae family, particularly the genera Proteus, Morganella, and Providencia, from other enteric bacteria.
