PHI 6 Agar is a specialized culture medium designed for use in the Gamma Phage Lysis Test, a confirmatory method used for identifying Bacillus anthracis. The medium supports the growth of the suspected organism while allowing interaction with the gamma bacteriophage, a virus that specifically infects and lyses B. anthracis cells. In the procedure, a bacterial lawn is prepared on the agar surface and a drop of gamma phage suspension is applied to a marked area. Following incubation, the presence of a clear zone or plaque indicates successful phage mediated lysis and confirms the identity of B. anthracis. The medium is used in diagnostic and reference laboratories for differentiating this organism from closely related Bacillus species.
