PHI X Agar is a specialized medium used for the cultivation and enumeration of the bacteriophage Phi X174, a virus that infects Escherichia coli. This bacteriophage is commonly used as a surrogate organism in filtration and viral removal studies because of its small and stability. The medium is frequently used in viral filtration efficiency testing of medical masks, respirators, and air filtration systems. The 12 mL plate format is designed to function as the base layer in the double layer agar technique, where a soft top agar containing host bacteria and the phage sample is poured over the surface. The bottom layer provides nutrients and structural support, allowing visible plaques to develop as the virus infects and lyses bacterial cells.
