Use platelet-derived growth factor (PDGF) BB to induce mesenchymal stem cell migration, angiogenesis, and osteogenesis (Xie et al. Nat Med, 2014). PDGF is a dimeric glycoprotein that can be assembled as heterodimers (PDGF-AB) or homodimers (PDGF-AA and PDGF-BB) (Westermark & Heldin. Acta Oncol, 1993). This glycoprotein signals through the receptor tyrosine kinases PDGFRα and PDGFRβ. PDGF is a potent mitogen for cells of mesenchymal origin and induces migration through signaling pathways involving MEK/ERK, EGFR, Src, and PI3K/AKT (Kim et al. Growth Factors, 2007). PDGF-BB is secreted by osteoblasts and preosteoclasts during bone modeling and remodeling (Xie et al.). Human recombinant PDGF-BB from STEMCELL is animal component-free and comes lyophilized with ≥ 95% purity; EC50 ≤ 20 ng/mL; endotoxin levels verified ≤ 1 EU/μg protein.
