Our Thermo Scientific 8-Well Strip Caps for Immuno Standard Modules were designed to provide a positive seal for flat and round bottom wells of our Nunc MicroWell 96-Well Microplates and Nunc Immuno 96-Well Plates.
8-Well Strip Caps for Immuno Standard Modules are not compatible with C or V bottom wells of the Nunc Microwell 96-Well Microplates and Nunc Immuno 96-Well Plates.

For Nunc Immuno plates and modules, what are the advantages of one well geometry type over another? Which types should I use for various applications?

The following list describes the geometries of wells available for Nunc Immuno-plates and modules:
- Flat bottom (F): Allows maximum transmission of light. These plates can be read on a monochromatic reader.
- Round bottom (U): This geometry optimizes washing and coating. These plates can be read using a dual wavelength reader.
- "C" bottom (C): This design of the well is a combination of both flat and round bottoms. Basically, it is a flat bottomed well with curved edges at the bottom. These plates also can be read using a monochromatic reader combining optimal reading and washing.
- StarWell: These wells have a modified "C" shape geometry with eight fins strategically placed at the bottom. This feature increases surface area, allowing more molecules to become immobilized which reduces incubation times.

Why am I getting a high background signal when using Nunc Immobilizer Streptavidin Plates/Strips?

High background signal could be due to inadequate washing. Make sure to pre-wash the Nunc Immobilizer Streptavidin Plates/Strips and increase the number of washing steps and the time between washes. Another possible cause is too high a concentration of the detection components.

How long will my Nunc Immobilizer Streptavidin Plates/Strips remain stable?

Our Nunc Immobilizer Streptavidin Plates/Strips are extremely stable over time, if stored at room temperature (20 degrees C to 25 degrees C). If kept in the unopened foil packaging, this product should have an 18 month shelf life after the date of manufacturing.

How long should I incubate the biotinylated and target molecule with the Nunc Immobilizer Streptavidin surface?

Coupling of the target molecule to the streptavidin proceeds very rapidly. Results show that incubation with gentle agitation for 2 hr at room temperature is typically sufficient to saturate all available active site.

What is the binding capacity for free biotin?

The total binding capacity for free biotin is:
- 20 pmol/well in a 96 well (coating volume 100 µL)
- 10 pmol/well in a 384 well (coating volume 50 µL)

Why should I pre-wash my Nunc Immobilizer Streptavidin?

We highly recommend pre-washing your Nunc Immobilizer Streptavidin. This will help ensure improved readouts and a very low coefficient of variation (CV% ≤ 5) from plate to plate and from well to well.

When using Nunc Immobilizer Streptavidin Plates/Strips for detection of biotinylated biomolecules, should I optimize the concentration of my target molecule?

We recommend that you optimize the amount of target molecule.
Typically, we suggest the following:
- Biotinylated oligonucleotides: 0.5 µM -0.01 µM diluted in 5 x SSCT buffer pH=7.0
- Biotinylated peptides: 1 µg/mL - 1 ng/mL diluted in a PBST buffer pH=7.2
- Biotinylated proteins: 5 µg/mL - 0.05 µg/mL diluted in a PBST buffer pH=7.2
