Description

Used to lyse cells under nondenaturing conditions. Invitrogen™ ProcartaPlex™ Cell Lysis Buffer is available in 20mL units that should be stored chilled (for continual use) or frozen (for longer periods of time.) Additional protease inhibitors can be added without any difficulties.

For lysis of adherent cells, 400μL of cell lysis buffer is used for a 10cm dish. For non-adherent cells, 400μL of buffer is used per 10⁷ cells, once they have been washed in PBS and pelleted. Buffer should be chilled and PMSF should be added to the buffer immediately prior to use.

Buffer composition:

• 20mM Tris-HCl (pH 7.5)
• 150mM NaCl
• 1mM Na2EDTA
• 1mM EGTA
• 1% Triton
• 2.5mM sodium pyrophosphate
• 1mM β-glycerophosphate
• 1mM Na3VO4
• 1μg/ml leupeptin
