Proguanil hydrochloride is an antimalarial proagent that is metabolized to its active form, Cycloguanil. It functions as a dihydrofolate reductase (DHFR) inhibitor. While Proguanil itself has weak antimalarial activity, its effectiveness is primarily attributed to Cycloguanil. The combination of Atovaquone and Proguanil shows synergistic activity in vitro, effective against gametocytes and pre-erythrocytic (hepatic) stages of malaria parasites. Additionally, Proguanil, its metabolite 4-chlorophenyl-1-biguanide (CPB), and Cycloguanil reversibly inhibit 5-HT3 receptor responses.
