What is the composition of the Protease Inhibitor Cocktail II after reconstitution?

When reconstituted, each vial of Protease Inhibitor Cocktail II contains the following components:

  • 20 mM AEBSF.HCl
  • 1.7 mM Bestatin
  • 200 µM E-64
  • 85 mM EDTA
  • 2 mM Pepstatin A

How can I reconstitute Protease Inhibitor Cocktail II?

Protease Inhibitor Cocktail II is recommended for use with bacterial cell extracts. You should first reconstitute each vial in 1 mL DMSO. Then add 4 mL DI water to obtain a 5 mL stock solution. The solution may appear turbid. The 5 mL stock is enough to inhibit protease activity extracted from a 20 g bacterial cell pellet.

How should I store reconstituted stock solutions of Protease Inhibitor Cocktail II?

After reconstitution, we recommend that you aliquot and freeze the solution at -20°C. Stock solutions are stable for at least 1 month at -20°C.

The Protease Inhibitor Cocktail II solution appears turbid after reconstitution. Is this normal and will the solution remain turbid or clear up?

If the solution appears turbid after reconstitution, the solution for this material will appear turbid throughout its usage. This is normal and the material will function without issue.

How do I prepare the Protease Inhibitor Cocktail II?

We recommend that you reconstitute each vial with 1 mL DMSO first, and then add 4 mL water to obtain a 5 mL stock solution.
