Pseudoprotodioscin is a furostanoside that inhibits SREBP1/2 and microRNA 33a/b levels, leading to a reduction in gene expression related to the synthesis of cholesterol and triglycerides. In Hep G2 cells, it increases ABCA1 protein and mRNA levels and promotes the efflux of ApoA-1-mediated cholesterol. It also inhibits SREBP1c and SREBP2 transcription by decreasing microRNA 33a/b levels, which reciprocally increases ABCA1 levels. In THP-1 macrophages, Pseudoprotodioscin reduces HMGCR, FAS, and ACC mRNA levels and promotes low-density lipoprotein receptor by decreasing PCSK9 levels.
