Puromycin dihydrochloride is an aminonucleoside antibiotic that inhibits protein synthesis. It blocks protein synthesis after aminoacyl-sRNA formation, leading to the accumulation of small peptides. It induces premature termination of translation by serving as an acceptor substrate and by competing with aminoacyl-tRNA for binding to the A' site. This allows for direct evaluation of translation activity in single cells using immunofluorescence microscopy and in heterogeneous cell populations via fluorescence-activated cell sorting.
