Puromycin Dihydrochloride is a potent antibiotic. It is an amino nucleoside antibiotic that mimics the 3'-end of aminoacyl-tRNA. It enters the ribosomal A site during translation, causing premature chain termination and inhibiting protein synthesis. Puromycin is incorporated into growing polypeptide chains, leading to the release of incomplete and nonfunctional peptides.
Widely used as a tool in cell biology to study protein synthesis and investigate translation processes. Its ability to terminate protein synthesis at the ribosome makes it valuable for various applications, including pulse-chase experiments and the analysis of nascent peptides. Puromycin resistance genes are often used as selectable markers in genetic engineering. Cells expressing the resistance gene can survive and proliferate in the presence of Puromycin, allowing for the selection of transfected or transduced cells.
