Purple Broth with Dextrose is a differential microbiological medium used to determine whether microorganisms can ferment the sugar dextrose (glucose). The medium contains bromocresol purple as the pH indicator and includes a Durham tube for detection of gas production. When bacteria ferment dextrose they produce acidic by products that lower the pH, changing the medium color from purple to yellow. If gas is produced during fermentation it becomes trapped inside the inverted Durham tube as a visible bubble. A yellow broth with gas indicates positive fermentation with gas formation, while yellow without gas indicates acid production only. If the medium remains purple, the organism does not ferment dextrose. This test is widely used in microbiology laboratories for bacterial identification and metabolic characterization.
