Purple Broth with Lactose is a differential culture medium used to evaluate an organism’s ability to ferment lactose. The formulation includes bromocresol purple as the pH indicator and a Durham tube for detection of gas production during carbohydrate metabolism. Bacteria capable of fermenting lactose break the sugar down into glucose and galactose, producing acidic by products that lower the pH of the medium. As acidity increases the indicator changes from purple to yellow, signaling a positive fermentation reaction. Gas formation during fermentation becomes trapped in the Durham tube as a visible bubble. This test is commonly used in microbiology laboratories to help differentiate lactose fermenting organisms such as Escherichia coli and Enterobacter from non fermenting species like Salmonella and Shigella.
