Purple Broth with Mannitol is a differential microbiological medium used to determine whether microorganisms can ferment the sugar alcohol mannitol. The medium contains bromocresol purple as a pH indicator and includes a Durham tube to detect gas production during fermentation. Organisms that metabolize mannitol produce acidic by products that lower the pH of the broth, changing the indicator color from purple to yellow. Gas generated during fermentation collects in the Durham tube as a visible bubble. A yellow broth with gas indicates positive fermentation with gas formation, while yellow without gas indicates acid production only. If the medium remains purple, the organism cannot ferment mannitol. This test is commonly used for differentiating bacterial species, particularly among Staphylococcus and certain Enterobacteriaceae.
